Metformin Usage

Metformin is a biguanide compound which originated from the extraction of goat’s rue plants. Metformin (Glucophage) was used in Europe for many years before it was approved for use in the United States in 1995. Metformin is the only approved biguanide for the treatment of patients with NIDDM that are refractory to dietary management alone. Metformin is quickly absorbed from the small intestine. Metformin remains the first-line pharmacological choice for the management of hyperglycaemia in T2DM. It works to decrease blood glucose during pregnancy.

Is it still worth pursuing the repurposing of metformin as a cancer therapeutic?

SR Lord, AL Harris

British Journal of Cancer volume 128, pages958–966 (2023)

This perspective article summarises how interest initially developed in the oncological potential of metformin and the diverse clinical programme of work to date including our contribution to establishing the intra-tumoral pharmacodynamic effects of metformin in the clinic. We also discuss the lessons that can be learnt from this experience and whether a further clinical investigation of metformin in cancer is warranted.

Mechanisms of cancer cell killing by metformin: A review on different cell death pathways

Xiao-yu Wu, Wen-Wen Xu, Xiang-kun Huan, Guan-nan Wu, Gang Li, Yu-Hong Zhou & Masoud Najafi

Molecular and Cellular Biochemistry volume 478, pages197–214 (2023)

We discuss the modulatory roles of metformin on different cancer cell death pathways including apoptosis, mitotic catastrophe, senescence, autophagy, ferroptosis and pyroptosis.
